I agree re: It's Complicated.
Also, have you seen Dirty Rotten Scoundrels with Steve Martin? It's not a slapstick, but there are a couple scenes that still bring tears to my eyes from laughing so hard, even after watching it 6+ times.

Today I was at walmart , and I saw a lady go by talking on her phone pushing a prob 1 yr old in the cart , she was just talking away, baby was going through her purse.
Wanna guess what he pulled out to chew on???

I like some mentioned already, yet absolutely felt I wasted an hour and a half of my life when I watched Nacho Libre (recommended by a friend who loved it - to each his own).
But not mentioned yet that I laugh with: SecondHand Lions, Gran Tornio (both movies are emotional, but the characters themselves have snappy comebacks and interesting personality).
If all else fails, turn on Funniest Home Videos. My husband, myself, and my two year old laugh the most together watching the show.

We love The Wedding Singer, Oh Brother Where Art Thou, Home Fries, The Long, Long Trailer (with Lucille Ball and Desi Arnaz), Pillow Talk with Doris Day and Rock Hudson.
I don't know if you can rent him on DVD, but Jim Gaffigan is an absulutely hilarious comedian and he's not dirty either. He's on Comedy Central a lot and we laugh our heads off everytime we watch him.
Laughter is good for the soul.
I hope you get some great responses.

Planes, trains and automobiles with John Candy and Steve Martin is a classic. If you need a good laugh check out http://awkwardfamilyphotos.com/ - photos people have submitted of family, kids, etc. that are just BAD - with hilarious captions. I looked at this this a.m. and literally was at my computer laughing until I cried!

Superbad was pretty funny. Spinal Tap is hilarious (as well as most of those Christopher Guest mockumentary movies). I also love anything with Ben Stiller (especially Dodgeball, for some reason, it cracks me up). You've gotten some really good suggestions already - I haven't seen all of them, so now I also have a list of movies to check out. Thanks for the question!

Okay, here's my favorite..... Mighty Aphrodite. It's a Woody Allen movie and the one that Mira Sorvino won her Oscar for, I think, in 1996. Her character has a fair amount of four-letter words in it, but not because she is a foul-mouthed woman just to swear. But she literally doesn't know any better. She is a big-hearted adult film actor who may possibly be the mother of the son that Woody Allen's character adopts with his wife, played by Helena Bonham Carter. The scene where these two meet is hysterical ("refund!" "refund!"). The rest of the movie has a bunch of funny stuff in it as well.
I actually used part of that movie for a dialect final in college (I was a theatre major). You'll recognize it if you see it, but it was the scene about the "onion farmer". Just picture it with a Swedish accent. Fun, fun, fun!
Happy laughing!
